Comparison of PBMCs between technologies
An early-access user was interested in improving efficiency and simplifying workflows using the CS Genetics SimpleCell™ Assay. Viably frozen PBMCs were processed with the SimpleCell™ 3′ Gene Expression Assay and with an alternative 3′ Gene Expression technology. Data from the two technologies were analysed separately and revealed the same cell type populations with relatively similar abundance. The data were then split by donor and integrated computationally. There was an excellent overlap between the data sets from the two technologies regardless of donor.
96 frozen whole blood samples
An early-access user was interested in a simplified high throughput single cell assay that works with frozen whole-blood samples to enable population scale single cell analysis. The CS Genetics SimpleCell™ Assay was used to process 96 frozen whole-blood samples in two days with the aid of a 96-channel semi-automated pipettor, thus showcasing the scalability of the technology. Gene expression quantification enabled high-resolution annotation of cell types within the samples.
iPSCs differentiation
An early-access user differentiated Human iPSCs to cortical neurons using an iNeuron protocol. Samples were collected in triplicate at 5 time points with a total of 15 processed for single cell RNA sequencing. Analysis of these combined datasets revealed distinct developmental states of the iPSCs during differentiation. Unsupervised pseudotime ordering revealed 4 key developmental stages, beginning with a formative stem-cell stage at D0, followed by a progenitor stage at D4. These were followed by more developmentally heterogenous stages up to D14, with cells existing in several pseudotime states (also potentially representing different developing neural types) while by D30 most cells reside in a mature state.
CAR-T cell screening comparison to an alternative single cell technology
Single cell sequencing can be used to find phenotypes of cells beyond what is possible with flow cytometry but, unfortunately, the cost with single cell technologies currently on the market can be prohibitively expensive for large scale applications like CAR-T donor screening. The CS Genetics SimpleCell™ Assay offers an easy, scalable and cost-effective solution for these applications with comparable performance to current technologies. An early-access user compared SimpleCell™ with another single cell technology and demonstrated that both datasets revealed comparable biological insights.
Quantification of a therapeutically-relevant gene-seq enrichment analysis (GSEA) signature demonstrated excellent correlation between the two technologies. The CS Genetics SimpleCell™ Assay showed similar quality, phenotypes and patterns in the transcriptional profiles.
